The insulin-like growth factor (IGF) system includes two peptide ligands, IGF-I and IGF-II; their receptors, the IGF-I receptor and the mannose-6-phosphate receptor, and six IGF-binding proteins. This family of growth factors plays fundamental roles in normal physiology and in neoplastic and metabolic disease states. Circulating IGFs are produced primarily in the liver but are also produced locally, resulting in both endocrine and autocrine/paracrine actions. Because of complexity in location and modes of regulation, previously unknown and important functions for the IGFs are rapidly being discovered.
