Over the years, activity-based protein profiling (ABPP) and bioimaging have been developed as powerful technologies for drug discovery. Specifically, both approaches can be applied in critical steps of drug development, such as therapy target discovery, high-throughput drug screening and target identification of bioactive molecules. In recent years, we have been focusing on developing various strategies that enable simultaneous activity-based protein profiling and bioimaging studies, thus facilitating an understanding of drug actions and potential toxicities. In this focus review, we summarized these novel strategies and applications, with the aim to promote these technologies in drug discovery.
